Application of a bioenergetic growth model to larvae of the pearl oyster Pinctada margaritifera L. ArchiMer
Thomas, Yoann; Garen, Pierre; Pouvreau, Stephane.
We applied, for the first time, a dynamic energy budget (DEB) growth model to the larval phase in the pearl oyster Pinctada margaritifera var. cumingii (Linnaeus, 1758) to evaluate the impact of spatio-temporal variation in the atoll lagoon environment on its capacity for development. The specific parameters of the model, which represent ingestion, temperature effect and the relationship between length and biovolume of the larvae, were determined from experiments or taken from the literature. The interpretation of the values of these parameters allowed us to identify the underlying adaptive character trait: P. margaritifera larvae have a good capacity to exploit low food concentrations and a narrow range of thermal tolerance restricted to hydrobiological...

Tahiti’s pearl farming industry plays a major socio-economic role in French Polynesia. In an increasingly competitive market where the production of high quality pearls becomes essential, research can help secure and ensure sustainable production. In that context, Ifremer, in close collaboration with the “direction des resources marines” (French Polynesian government agency) has developed research projects on the “sustainable development of pearl farming”. This program is organized along 3 axes: (1) understanding the animal physiology and initiating a genetically selective breeding program of donor oysters; (2) understanding pearl oyster larvae dispersal and recruitment; (3) understanding the molecular mechanisms underlying biomineralization processes...

Comparison of growth of mussel, Mytilus edulis, on longline, pole and bottom culture sites in the Pertuis Breton, France ArchiMer
Garen, Pierre; Robert, Stephane; Bougrier, Serge.
Growth of the mussel, Mytilus edulis, was compared for the first time under three culture regimes, longline located in open sea, pole and on-bottom both situated on an intertidal flat, in the Pertuis Breton, on the Atlantic coastline of France. Mussel sampling was performed on a monthly basis over 1 year while monitoring of hydrobiological parameters was conducted on a biweekly basis. Fluctuation of environmental parameters showed a similar pattern on both sea and shore locations with generally higher concentration levels for the intertidal area compared with the open sea, i.e. 4.07 and 3.17 mg 1(-1) for particulate organic matter, 3.16 and 2 mug 1(-1) for chlorophyll-a, 58.43 and 38.72 mg 1(-1) for inorganic-N, respectively. A clear seasonal growth...

Continuous fluorescence recording as a way to improve Pacific oyster (Crassostrea gigas) models of paralytic shellfish toxin accumulation ArchiMer
Baron, Regis; Couedel, Marielle; Joret, Camille; Garen, Pierre; Truquet, Philippe; Masselin, Pierre; Bardouil, Michele; Lassus, Patrick.
A simple system was used to simulate the effect of alternating toxic (paralytic shellfish poisoning toxins) and non-toxic microalgal diets on oyster feeding behaviors and rates of toxin accumulation. These experimental conditions were meant to reflect, to some extent, the incoming and outgoing fluxes of toxic algae observed at the mouth of the Penze estuary (Northern Brittany, France). Physiological and toxicological parameters were estimated based oil fluorescence measurements recorded continuously at the outlet of each experimental tank, which contained a single oyster. Q(tox), this variable describes toxin uptake in oysters, it was used (instead of the toxin ingestion rate): i) in simple graphical analyses, ii) as well as in one- and two-compartment...

Development of in situ hybridisation using 16S rRNA gene to monitor black-lip pearl oyster, Pinctada margaritifera, larvae in plankton samples ArchiMer
Thomas, Yoann; Belliard, Corinne; Garen, Pierre; Gueguen, Yannick; Montagnani, Caroline.
In French Polynesia, the black-lip pearl oyster Pinctada margaritifera has been farmed to produce pearls since the 1980s, forming the basis of a major industry. The sustainability of this activity relies on spat collection in the lagoons. However, pearl oyster spat can be difficult to identify for the evaluation of stock variations. It is especially hard to distinguish Pinctada spp. larvae at a very early stage of development. In the present study, a whole- mount in situ hybridisation (ISH) technique was developed to allow the discrimination of larvae of closely-related pearl oyster species found in the French Polynesian atolls. Using specific ribosomal 16S-DNA sequence data, we were able to successfully differentiate between Pinctada margaritifera and...

Multi-scale distribution and dynamics of bivalve larvae in a deep atoll lagoon (Ahe, French Polynesia) ArchiMer
Thomas, Yoann; Garen, Pierre; Bennett, Auguste; Le Pennec, Marcel; Clavier, Jacques.
Bivalve larvae and hydrographic parameters were sampled over a range of spatio-temporal scales in a deep atoll lagoon. Bivalve larvae abundances were very high throughout the year: 18,550 m(-3) in average. Larvae were (i) concentrated at mid-depth with nocturnal ascent and diurnal descent, (ii) heterogeneously dispersed at the lagoon scale, (iii) subject to day-to-day variation in abundance and (iv) transferred between different parts of the lagoon providing evidence of intra-lagoonal connectivity. The primacy of physical factors was seen on large spatial scale with the diluting effect of water renewal and transfers by hydrodynamics. On smaller spatial scale, the primacy of biological processes was recognised, with larval swimming activity leading to dial...

Paralytic shellfish poison outbreaks in the Penze estuary: Environmental factors affecting toxin uptake in the oyster, Crassostrea gigas ArchiMer
Lassus, Patrick; Baron, Regis; Garen, Pierre; Truquet, Philippe; Masselin, Pierre; Bardouil, Michele; Leguay, Didier; Amzil, Zouher.
Several experiments using a self-regulated system were conducted to define the factors likely to influence the uptake of paralytic shellfish poison (PSP) by oysters in the Penze estuary (France, Brittany). Each 4-day experiment was carried out in a recirculated sea water system using 15 Pacific oysters (Crassostrea gigas) separated from each other and supplied with unfiltered natural seawater containing alternatively toxic (Alexandrium minutum) or non-toxic (Skeletonema costatum) algal diets. The food supply and exposure times to toxic diets were determined according to field studies of the Upstream and downstream movement of patches containing A. minutum. The experimental parameters corresponded roughly to the hydrological conditions generally observed in...

Response of the pearl oyster Pinctada margaritifera to cadmium and chromium: Identification of molecular biomarkers ArchiMer
Gueguen, Yannick; Saulnier, Denis; Santini, Adrien; Magre, Kevin; Garen, Pierre; Bernagout, Solene; Nohl, Marine; Bouisset, Patrick; Helme, Herehia; Planes, Serge; Le Moullac, Gilles.
This study was designed to identify in the pearl oyster Pinctada margaritifera, used as a bio-accumulator, molecular biomarkers for the presence of heavy metals in the lagoon environment. Pearl oysters were exposed to 2 concentrations (1 and 10 μg L− 1) of cadmium (Cd) and chromium (Cr) compared to a control. Twelve target genes encoding proteins potentially involved in the response to heavy metal contamination with antioxidant, detoxification or apoptosis activities were selected. P. margaritifera accumulated Cd but not Cr, and mortality was related to the amount of Cd accumulated in tissues. In response to Cd-Cr contamination, metallothionein (MT) was significantly up-regulated by Cd-Cr at both concentrations, while 7 others (SOD, CAT, GPX, GSTO, GSTM,...

Spatial and temporal variability of the pico- and nanophytoplankton and bacterioplankton in a deep Polynesian atoll lagoon ArchiMer
Thomas, Y.; Garen, Pierre; Courties, C.; Charpy, L..
Spatial and temporal scales of variability in phyto- and bacterioplankton abundance were investigated in a deep atoll lagoon (Ahe, French Polynesia), Ahe Lagoon showed a classical phytoplankton composition, but its picophytoplankton (Prochlorococcus, Synechococcus and picoeukaryotes) concentrations are higher than the mean for Polynesian atoll lagoons. Phyto- and bacterioplankton descriptors were highly variable, with mean CVs of >20% in most cases: day night, day-to-day, vertical, lagoonal and seasonal. Phytoplankton biomass, which was estimated by in vivo fluorescence, was higher in the deep layer than in the upper layer. This pattern was detected on a day night scale, showing transitory vertical gradients, and through long-term events that were...

Test du potentiel antisalissure de certaines macromolécules exopolysaccharides pour l'élevage d'huîtres perlières ArchiMer
Garen, Pierre; Tiapari, Jerome.

The limits of morphometric features for the identification of black-lip pearl oyster (Pinctada margaritifera) larvae ArchiMer
Paugam, A; D' Ollone, Christine; Cochard, Jean-claude; Garen, Pierre; Le Pennec, Marcel.
As with most cultivated bivalves, culture of the Tahitian pearl oyster Pinctada margaritifera is particularly dependent on the natural environment, especially for spat supply. The ability to track in real time the abundance and the development of pearl oyster larvae in the plankton would help optimize spat collection in atolls of French Polynesia. However no identification criteria are available for the larvae of several bivalves species present in the lagoons and it is not yet possible to specifically monitor pearl oyster larvae. The aim of this study is to determine the most pertinent morphological identification criteria, to specifically identify the larvae of P. margaritifera and differentiate them from those of three other abundant species: Pinctada...
